Stephenie Meyer-The Twilight Series
Stephenie Meyer posted on her Q.A. section of her fan site that she didn't know anything about publishing but she just knew she had a book that she wanted others to read. She used google to search for how to publish and subscribed to WritersMarket.com. Stephenie got seven or eight rejection letters before being accepted. Her advice for others wanting to be published is to finish the writing completely, edit it, then find agents using Writer's Market. After that, research those agents to see what type of a query they want and send out plenty of query letters. She also says to expect rejection letters but don't get discouraged.
J.K. Rowling-The Harry Potter Series
J.K. Rowling was able to get her first book published by going through an agent. She was turned down numerous times until one accepted her work. Her advice on publishing is: "I would advise any novice to get one [an agent] if they can. Other than your agent, you deal with an editor and perhaps the people marketing your book. Most of the time it takes more than just the first agency to accept an author's work. it is important to send to more than one in order to have a better chance to become published.

Barbara Kingsolver
Barbara Kingsolver had an agent and says that every author aspiring to get published needs. Many publishers won't even give a look to anything without an agent. She started with writing small articles and journals in newspapers and then on to novels.
